Molecular Mechanisms of Cardioprotective Effects
Apigenin exhibits a wide range of biological activities, including antioxidant, anti-inflammatory, anticancer, and neuroprotective properties. Studies demonstrate that apigenin (125 mg/kg and 250 mg/kg) mitigates doxorubicin (ADR)-induced myocardial injury (24 mg/kg) by suppressing serum aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) release, while reducing creatine kinase (CK) levels.
Additionally, apigenin inhibits cytochrome P450 2C9 (CYP2C9) in the CYP2C9 RECO system (a recombinant enzyme system containing human CYP2C9, P450 reductase, cytochrome b5, and liposomes) with a Ki of 2 μM. It also suppresses cell proliferation: after 7 days, 20, 40, and 80 μM apigenin achieved growth inhibition rates (IR) of 38%, 71%, and 99%, respectively. Clonal formation of SGC-7901 cells was dose- and time-dependently inhibited after 24 or 48 hours of exposure. At 80 μM, cloning efficiencies dropped to 9.8% (24h) and 5% (48h), compared to 40.4% and 43.4% in control groups.
